The nucleic acid detection of the corona virus is the key process to the diagnosis. The detection procedure needs to go through five steps: Pcr Nasal Swab sampling, sample retention, viral transport medium sample preservation, nucleic acid extraction, and nucleic acid Amplification Test.
Step 1, Pcr Nasal Swab Sampling
Carefully insert the nasopharyngeal swab into one nostril. Using a gentle rotation push the swab up to 25 cm(1inch) from the edge of the nostril. Rotate the nasopharyngeal swab 5 times against the mucosa inside the nostril to ensure sufficient specimen collecction.
Using the nasopharyngeal swab repeat this process in the other nostril to ensure that the adeguate amount of sample is collected from both nasal cavitiess. Withdraw the nasopharyngeal swab from the nasal cavity.
Step 2, Sample Retention
This step requires professional trained healthcare personnel to retain samples, immerse the Pcr Nasal Swab/ Nasopharyngeal swab head in the viral transport medium and immediately tighten the cap of viral transport medium tube after breaking the tail.
Step 3, sample preservation
Put the viral transport medium tube into a sealed bag and send it for inspection in time. During the inspection process, a strict transportation environment is required, and it is stored at 2-8 degrees Celsius.
Step 4, nucleic acid extraction
Extract the nucleic acid from the virus-inactivated sample for subsequent detection. Automated equipment, such as a nucleic acid extractor, can be used.
Step 5, nucleic acid Amplification Test
After the extract is fluorescent PCR amplification reaction, which takes 70-80 minutes.
